Lipid Extraction
Lipidomics analysis benefits from extraction with nonpolar organic solvents since polar metabolites, proteins and salts were reduced, leading to a greatly reduced complexity of the samples and MS readout.
- Procedures need to be evaluated concerning recovery and reproducibility of measured lipids for respective sample matrix
- Exogenous (it should be verified that they are not present in the sample) internal standards should be added prior to extraction; equilibration with the sample should be evaluated and also verified not present in the sample
- Frequently applied protocols:
- Bligh and Dyer
- Folch
- MTBE
- 2-phase BUME for plasma and tissue
